Abstract
This study was to investigate the geochemistry and the indigenous acidophilic bacteria in the acid mine drainage from the abandoned Sama-Taejeong coal mine, Gangwon-do. The size of the rod-shaped bacteria observed in the acid mine drainage ranged from 0.3×1.3 ㎛ to 0.4×3.4 ㎛, and sheathed-filament bacteria were observed in the yellow-colored iron hydroxide. These rod-shaped indigenous bacteria were grown in a growth- medium containing elemental sulfur and Fe2+ ion. As the rod-shaped bacteria were growing, the pH value was decreased from 3.72 to 2.08, whereas the Eh values were increased in the growth-medium. In decreasing the pH value and increasing the Eh value in the growth-medium and the subculture, it was identified that the rod-shaped indigenous bacteria were utilized the Fe2+ and elemental sulfur. The zero point of charge of the yellow-colored iron hydroxide were determined to range from 4.1 to 7.1 using zeta potential.
도계 삼마-태정 폐탄광에 형성된 산성광산배수의 지구화학적 특성과 광산배수 중에 서식하는 토착박테리아의 존재를 조사하고자하였다. 크기가 0.3×1.3 ㎛에서 0.4×3.4 ㎛ 범위인 막대 모양의 박테리아들이 산성광산배수에서 관찰되었으며 협막을 형성하는 박테리아는 황갈색 철수산화물에서 관찰되었다. 이들 막대 모양의 토착박테리아들이 원소 황과 Fe2+ 이온이 포함된 성장-배양액에서 성장되었다. 막대 모양의 박테리아들이 성장함에 따라 성장-배양액의 pH는 3.72에서 2.08로 감소하였고, Eh는 증가하였다. 성장-배양액과 계대배양에서 이들 막대 모양의 토착박테리아들이 배양됨에 따라 이들 박테리아들은 Fe2+ 이온과 원소 황을 활용하는 박테리아들임이 밝혀졌다. 성장-배양액과 계대배양에서 감소하는 pH 값과 증가하는 Eh 값은 막대 모양의 토착박테리아들이 Fe2+ 이온과 원소 황을 활용하고 있음이 확인되었다. 황갈색 철수산화물에 대한 제타전위를 측정한 결과 등전위가 4.1에서 7.1로 나타났다.
